The novel is divided into three parts, each corresponding to a different historical era. The first part, titled "Ang Paglalakbay", covers Simon's childhood and youth under the Spanish rule. He witnesses the abuses and injustices committed by the friars and the guardia civil, as well as the resistance and rebellion of his fellow Filipinos. He also falls in love with Maria Clara, a mestiza who is betrothed to a Spanish officer. The second part, titled "Ang Pagbabalik", follows Simon's return to San Roque after spending several years in Manila and abroad. He becomes a wealthy businessman and a leader of the local community. He also reunites with Maria Clara, who is now a widow with a son. However, he faces new challenges and enemies, such as the Japanese invaders and the corrupt politicians. The third part, titled "Ang Pagtutuos", depicts Simon's final confrontation with his nemesis, Don Pedro, a former guardia civil who has become a powerful landlord. Simon leads a peasant uprising against Don Pedro's tyranny, while also dealing with his personal conflicts and tragedies.

Dugo sa Bukang Liwayway is considered one of the first Philippine novels written in Filipino language. It uses an impeccable language style that blends poetic and realistic elements. It also addresses social problems, such as poverty, oppression, corruption, and violence, that are still relevant today. It reflects a part of Filipino culture, such as the values of family, faith, and patriotism. It also helps in the creation of the sense of identity of Filipinos, as it portrays their struggles and aspirations throughout history.
